PROJECT SUMMARY / ABSTRACT Coronary heart disease (CHD) remains the leading cause of death, with blood lipid levels representing critical, modifiable risk factors. This R01 renewal proposal by the Global Lipids Genetic Consortium (GLGC) seeks to advance understanding of how genetic variations influence lipid levels and CHD across all individuals and life stages, with a view toward refining risk assessment and preventive strategies. In Aim 1, we will identify genetic variants with age-specific effects on lipid levels by analyzing an expanded cohort of 2.5 million participants. We expect this approach will capture changes in lipid-related genetic influences at different ages. In Aim 2, we will address the genetics underlying lipid progression and CHD recurrence, deploying genome-wide analyses to uncover novel genetic markers that predict disease progression. In Aim 3, we will develop a new clinical polygenic score assay optimized for young adults (18-40 years) toward improving CHD prevention. This proposal is a renewal of the previous GLGC award and established collaborative relationships and will allow for the continued effort and coordination of the consortium with new clinical tra…
