Research Focus
Project Summary/Abstract Advances in molecular ‘omics measurement over the last decade made researchers’ access to rich unbiased datasets routine. However, an overarching limitation remains - to analyze the molecular contents of cells at high multiplexity (ie ‘omics approaches) requires destruction of the cells under study, either by lysis and extraction of the target analyte class, or by killing/fixation to introduce detection reagents. This prevents serial measurement of living cells to monitor dynamic processes. The need for time-dependent ‘omics measurements is made clear by multiple lines of evidence. 1) The proliferation of dozens of ‘pseudo-time’ analysis algorithms that infer time-dependent molecular evolution of biological systems with ‘omic breadth and 2) the pervasive impact of fluorescent protein technology for monitoring live-cell dynamics (one or a few molecules at a time) across biomedical research. Researchers now need tools that can read out living biology at ‘omic scale to follow complex dynamics and responses over time, and ultimately to bring such studies to living cells in live animals that cannot be accessed repeatedly or at all by existing ‘omics approaches.…
