Research Focus
Project Summary Our lab works in the general area of computational structural biology although it also includes an experimental component. We carry out theoretical and computational research and develop software tools, with our efforts being guided by a variety of applications of biomedical importance. We currently have research programs in three distinct areas all involving protein-protein interactions (PPIs): 1) The structure informed proteome-wide prediction of PPIs and their application to the discovery of PPI networks which are used to assign function at different levels of granularity. 2) How PPIs control cell-cell recognition and, specifically, neuron-neuron recognition. This work has a major experimental component involving binding affinity measurements of designed mutants. 3) The use of Free Energy Perturbation (FEP) calculations to guide the design of antibodies with improved binding properties. The overarching goal of the current proposal is to provide a structure-informed perspective in multiple areas of systems biology thus filling a major gap in this important and rapidly growing area of biomedical research. Although much experimental and computational progress has be…
