Research Focus
Project Summary/Abstract Our laboratory is recognized as a leader in bacterial epigenomics and high-resolution microbiome studies. Over the past decade, we have pioneered the use of single-molecule long-read sequencing technologies to comprehensively map bacterial DNA methylation events. This work has catalyzed a global effort, leading to over 5,000 bacterial epigenomes mapped to date and uncovering significant insights into the epigenetic mechanisms bacteria use to adapt to various stresses and host environments. We have also expanded our research from individual bacteria to microbiomes, introducing bacterial DNA methylation as epigenetic barcodes for high-resolution microbiome analysis. This innovative approach allows for strain-level resolution of metagenomic contigs and precise linkage of mobile genetic elements, such as plasmids, to their host bacteria. Our long-term goal is to thoroughly understand epigenetic regulation in bacteria and the microbiome to significantly impact biomedicine, systems and synthetic biology, and microbiome research. In the past five years, we have achieved substantial progress in developing methods and gaining biological insights into high- resolutio…
