Research Focus
PROJECT SUMMARY/ABSTRACT Research in the Kleiner group focuses on understanding molecular mechanisms of post-transcriptional RNA regulation. RNA regulation plays a central role in gene expression and aberrations in this process are implicated in human disease. Further, therapeutic agents that leverage understanding of RNA regulatory mechanisms are in clinical use and development. Major post-transcriptional regulatory modalities include post-transcriptional (or epitranscriptomic) modifications on RNA installed by cellular enzymes, interactions of RNA with RNA-binding proteins, and RNA-small molecule interactions. The Kleiner group leverages chemical biology approaches and proteomic/transcriptomic methods to investigate these processes in their native biological contexts. Mapping RNA modifications and characterizing their function is an outstanding challenge in biology with major implications for human health. Previous work in the group has used chemical probes, including modified nucleosides and oligonucleotides, to study RNA modification reader and writer enzymes. We have also developed and applied RNA mass spectrometry platforms and modification-specific chemistries to quantitativ…
