Research Focus
PROJECT SUMMARY Fluorescence microscopy has transformed biological research, enabling super-resolution imaging of biomolecular complexes and advancements in spatial transcriptomics and proteomics. These methods have provided critical insights into the localization and interactions of DNA, RNA and proteins, while deep learning has enhanced the analysis of large microscopy datasets. However, most of these achievements are based on fixed samples, overlooking the dynamic behavior of biomolecules and organelles in live cells. Live-cell imaging faces significant challenges, including photobleaching, phototoxicity, and technical limitations such as high cost, low throughput, and insufficient detection sensitivity. To overcome these barriers, this research proposes the development of innovative 5D subcellular imaging tools that integrate advanced optics, microscopy, photophysics, and deep learning-based analysis. The objective is to enable live-cell imaging with high spatial and temporal resolution, extended observation times, and minimal photodamage. These advancements are designed to transform live-cell imaging, facilitating the study of subcellular structures and biochemical processes i…
