Research Focus
Project Summary Drug-based therapies often suffer from inter-individual variability in drug metabolism, leading to suboptimal treatment outcomes. Cytochrome P450 drug-metabolizing enzymes (dmCYPs) metabolize nearly 70% of all clinically used medications. Of these, eleven dmCYPs (CYP1A2, 2A6, 2B6, 2C8, 2C9, 2C19, 2D6, 2E1, 3A4, 3A5, and 3A7) are of key clinical importance. Genetic variation in these genes significantly affects drug metabolism and forms the basis of pharmacogenetic (PGx) biomarkers used to guide therapy. However, current PGx panels have limited predictive accuracy due to two major challenges: (1) the principles governing inter- individual dmCYP expression remain poorly understood due to technical and biological complexity, and (2) existing panels are primarily optimized for individuals of European ancestry, limiting their clinical utility across different populations. The overarching goal of this research program is to identify all determinants of dmCYP variability across ancestral groups to inform the development of accurate PGx panels for personalized drug therapy for all populations. My team is uniquely positioned to achieve this goal. We have nearly two decades o…
