ABSTRACT – CORE B – BIOINFORMATICS AND IMMUNOLOGY Immunotherapies such as immune checkpoint blockade (ICB) and CAR-T have been successful in treating cancers. The most common brain tumor, glioblastoma (GBM), is highly recurrent and has poor prognosis even with multimodality treatment including surgical resection, radiation therapy, and chemotherapy. Although GBM is deemed immunologically “cold”, immunotherapies, including adoptive immune cell transfer therapy, is promising to achieve desired efficacy and safety for GBM treatment. The three RPs proposed in this P01 aim to study how novel polymer-based GBM vaccine (RP1), in vivo generation of CAR-T using a novel biomaterial (RP2), novel CAR-NKT therapy (RP3), and their combinations can benefit GBM treatment in various pre-clinical GMB animal models (Core C). Rational and successful development of effective cancer immunotherapies depends on comprehensive and in-depth immunological evaluations of tumor microenvironment (TMiE) and tumor macroenvironment (TMaE) during GBM development and its treatment. These evaluations are important to not only identify meaningful immunological parameters correlating with GBM development and effective t…
